| In vitro study | Pemetrexed effectively inhibits thymidylate synthase (TS) with K I of 1.3 nM and also significantly inhibits other key folate enzymes, including dihydrofolate reductase (DHFR) and glycinamide nucleotide carbamoyltransferase (GARFT) , K I was 7.2 nM and 65 nM, respectively. Pemetrexed has antiproliferative activity in CCRF-CEM leukemia, GC3/C1 colon cancer, and HCT-8 ileocecal cancer cells with IC50 of 25 nM, 34 nM, and 220 nM, respectively. Moreover, the combination of thymidine and hypoxanthine on the above three cell lines can completely reverse the cytotoxicity caused by ly231514. Recent studies have shown that the combination of Cisplatin and Pemetrexed has anti-cancer effects by inhibiting cell proliferation, invasion, and inducing apoptosis in MPM cells infected with Adenovirus of the vectors expressing SOCS-1. |
| In vivo study | Pemetrexed acts on human H460 non-small cell lung cancer xenograft, delaying tumor growth. Furthermore, Pemetrexed in combination with paclitaxel delayed H460 tumor growth to a greater extent and in addition to Vinorelbine and Carboplatin. |
